چکیده
Identification of molecular determinants receptor-ligand binding could significantly increase the quality structure-based virtual screening protocols. In turn, drug design process, especially fragment-based approaches, benefit from knowledge. Retrospective campaigns by employing AutoDock Vina followed protein-ligand interaction fingerprinting (PLIF) identification using recently published PyPLIF HIPPOS were main techniques used here. The ligands and decoys datasets enhanced version database useful (DUDE) targeting human G protein-coupled receptors (GPCRs) employed in this research since mutation data are available be to retrospectively verify prediction. results show that method presented article pinpoint some verified determinants. is therefore suggested as a routine discovery.

Molecular determinants of ligand binding to H4R species variants.
The histamine H(4) receptor (H(4)R) is the latest identified histamine receptor to emerge as a potential drug target for inflammatory diseases. Animal models are employed to validate this potential drug target. Concomitantly, various H(4)R orthologs have been cloned, including the human, mouse, rat, guinea pig, monkey, pig, and dog H(4)Rs. In this article, we expressed all these H(4)R orthologs...
متن کاملMolecular determinants of differential ligand sensitivities of insect ecdysteroid receptors.
The functional receptor for insect ecdysteroid hormones is a heterodimer consisting of two nuclear hormone receptors, ecdysteroid receptor (EcR) and the retinoid X receptor homologue Ultraspiracle (USP). Although ecdysone is commonly thought to be a hormone precursor and 20-hydroxyecdysone (20E), the physiologically active steroid, little is known about the relative activity of ecdysteroids in ...
متن کاملThe Molecular Determinants of Small-Molecule Ligand Binding at P2X Receptors
P2X receptors are trimeric eukaryotic ATP-gated cation channels. Extracellular ATP-their physiological ligand-is released as a neurotransmitter and in conditions of cell damage such as inflammation, and substantial evidence implicates P2X receptors in diseases including neuropathic pain, cancer, and arthritis. In 2009, the first P2X crystal structure, Danio rerio P2X4 in the apo- state, was pub...
متن کاملMapping the ligand binding sites of kainate receptors: molecular determinants of subunit-selective binding of the antagonist [3H]UBP310.
Kainate receptors (KARs) modulate synaptic transmission and plasticity, and their dysfunction has been linked to several disease states such as epilepsy and chronic pain. KARs are tetramers formed from five different subunits. GluK1-3 are low affinity kainate binding subunits, whereas GluK4/5 bind kainate with high affinity. A number of these subunits can be present in any given cell type, and ...
